Preparing for a sample
Turn on all components of the fluorometer, which is the stack of boxes/power supplies to the right and slightly behind the fluorometer.
- Select B on the data switch for Fluorescence spectroscopy.
To allow the light to pass to the light source find a black “shutter” between the two halves of the PFI fluorometer. At the top of the shutter is a double ended arrow labeled open and closed. Slide the black metal shutter toward you.
Select the appropriate signal multiplier, using the black knob under the multiplier. When the white stripe is facing the user it is on. When facing the 4 o’clock position it is not selected.
- Log on to the PTI Felix 32 Software, the password can be found on the manual cover. The manual is located in the pull out drawer to the right of the monitor. (Use the computer to the right of the fluorometer)
- Select appropriate multiplier. The dial is under
Running a sample
- Place sample in clean quartz cuvette. Students in CH332 will find the class’s cuvettes in the pull out drawer labeled 332, which is under the chalk board.
- Lift lid on the top of the fluorometer. The lid opens from the right and hinges at the left like a book.
- Place sample in cuvette holder and close lid. The lid will hold it self open unless you push to the right on the hinge to disengage the catch that keeps it open.

- Select Acquisitions, Open Acquisitions for current methods or New Acquisitions for creating a new method.
- If you have chosen New Acquisitions, choose the type of scan to be used, for example emission/excitation or timebase.
- Select wavelengths of excitation and emission, select step time and integration times, or points per second and duration. These values can be saved to reuse on other scans.
- Select Aquire(prep), you may need to wait a moment.
- When software is ready press Start.
- Your emission scan will be visible on the screen and can be saved by selecting Save from the menu.
- Remove cuvette and shut down power supplies: the order in which they are turned off is not important.
